In order to examine how Notch signaling maintains renal epithelia in a monolayer during renal proximal tubule morphogenesis we are utilizing an in vitro cell culture system that involves growing wild type and Notch signaling deficient renal epithelial cells in embedded extracellular matrix. This results in wild type renal epithelial cells forming a spherical structure composed of an epithelial monolayer with a single lumen. This video consists of serial confocal images of a single sphere stained for actin filaments to reveal the shape of each epithelial cell within the sphere, the presence a single lumen and absence of epithelial stratification.
